Sperm DNA fragmentation is a significant factor in male infertility despite having a normal sperm count in semen. It can be caused by various factors such as aging, chemotherapy, genetics, smoking, stress, and more. Currently, assessing sperm DNA fragmentation is one of the diagnostic tests to determine the cause of male infertility. There are different methods for this test, including TUNEL (Terminal deoxynucleotidyl transferase dUTP nick end labeling), SCSA (Sperm Chromatin Structure Assay), and COMET (Single Cell Gel Electrophoresis). The disadvantages of these methods are: 1) Complex procedures, 2) Need for expensive equipment, 3) Production by foreign companies, 4) Requirement for experienced personnel to interpret results, 5) Time-consuming.
Therefore, we aimed to design a kit that overcomes these disadvantages and can compete with foreign products by localizing this test. The principle of this kit is based on the fact that normal sperm contains more protamine and less histone compared to sperm with abnormal DNA. This allows the DNA of normal sperm to unwind easily in the presence of reducing solutions, forming a halo around the sperm head. In contrast, the DNA of abnormal sperm does not unwind and remains compact, not forming a halo around the sperm head.
